In Vivo 31P-Nuclear Magnetic Resonance Studies on Energy Metabolism of Ischemic Gerbil Brain: Comparison between Unilateral and Bilateral Common Carotid Artery Occlusion Models

چکیده
The changes of cerebral energy state under bilateral and unilateral common carotid artery occlusion-reflow models in Mongolian gerbils were studied using in vivo 31 P nuclear magnetic resonance spectrometry (3IP-MRS). All ten gerbils used in the bilateral common carotid artery occlusion-reflow model showed relatively uniform ischemic patterns, i.e., depletion of phosphocreatine (PCr) and adenosine triphosphate (ATP) and increase of inorganic phosphate (Pi) followed by relatively uniform recovery pattern of PCr, ATP and Pi. Among 20 gerbils used in the unilateral common carotid artery occlusion-reflow model, 6 animals displayed the ischemic pattern but 14 animals showed no significant changes during ischemia. The ischemic pattern recovered more rapidly in the unilateral ischemic group than in the bilateral one. Although the Mongolian gerbil has used customarily in the study of brain ischemia, the unilateral common carotid artery occlusion model showed significant interindividual variation concerning the change of brain energy state during ischemia. While the results of this study are essentially predictable from the prior works using classic biochemical methods, they are useful in the sense that we demonstrate the complete time course of changes in metabolites in the same animal.

A 31P nuclear magnetic resonance in vivo study of cerebral ischaemia in the gerbil.
We have used the noninvasive method of 31phosphorus nuclear magnetic resonance (31P NMR) in vivo to follow changes in phosphorous metabolite concentrations and the intracellular pH in the right and left hemispheres and in the cerebellum of gerbil brains after the occlusion of the right carotid artery. Spatial resolution over the brain was possible using surface coils. Ligation, which is know to...
متن کاملCell proliferation after ischemic infarction in gerbil brain.
In order to study cell proliferation after ischemic infarction, a model of bilateral common carotid artery occlusion in the gerbil was developed. A comparison of survival rates after 15, 30, 45 and 60 min of occlusion revealed that 45 min was the maximum duration of ischemia after which most (72%) of the gerbils were alive at 1 week. The administration of pentobarbital (single dose, 30 mg/kg) p...
متن کاملThe Effects of Sevoflurane on Recovery of Brain Energy Metabolism After Cerebral lschemia in the Rat: A Comparison with lsoflurane and Halothane
Isoflurane is an appropriate anesthetic for neuroanesthesia. We evaluated whether the effect of sevoflurane is similar to that of isoflurane or halothane on brain energy metabolism after cerebral ischemia followed by reperfusion using 31P-magnetic resonance spectroscopy. Wistar rats (n = 21) were divided into three groups: isoflurane-, sevoflurane-, or halothane-treated. After anesthesia induct...
متن کاملRegional cerebral glucose metabolism during and after bilateral cerebral ischemia in the gerbil.
Cerebral metabolic rate for glucose (CMRG) was measured using the 14C-deoxyglucose technique in a stroke model of the gerbil produced by bilateral common carotid artery occlusion. During 30 minutes of ischemia, 14C-deoxyglucose uptake in the brain was increased along the border zone between the ischemic and nonischemic area and decreased in the ischemic areas. During the early stage of reperfus...

BACKGROUND AND PURPOSE Diffusion-weighted magnetic resonance imaging has been shown to be particularly suited to the study of the acute phase of cerebral ischemia in animal models. The studies reported in this paper were undertaken to determine whether this technique is sensitive to the known ischemic thresholds for cerebral tissue energy failure and disturbance of membrane ion gradients.
